(The Profile of Immune Response Against Newcastle Disease in Broiler were Vaccinated with IBD-Inactivated Vaccine and Challenged by IBD Virus)

ABSTRAK

Penelitian ini bertujuan untuk mengetahui respon kebal terhadap Newcastle Disease pada ayam pedaging (broiler) yang divaksin IBD-inaktif dan ditantang dengan virus IBD. Sebanyak 200 ekor ayam pedaging strain Cobb dibagi menjadi empat kelompok (K1, K2, K3 dan K4). Vaksinasi dilakukan pada hari ke-4, dimana kelompok K1 dan K2 divaksin dengan vaksin ND aktif dan ITA ND+IBD inaktif, sedangkan kelompok K3 dan K4 hanya divaksin dengan vaksin ND aktif dan ND inaktif. Uji tantang dengan virus IBD dilakukan pada hari ke-14, terhadap kelompok K1 dan K3. Pada hari ke-28 dilakukan vaksinasi ulang ND (booster) terhadap semua kelompok. Sampel yang diambil berupa darah untuk pengukuran titer antibodi ND dan dilakukan pada hari ke-4 (antibodi maternal), 14, 21, 28 dan 42. Pengukuran titer antibodi ND dilakukan dengan uji hemaglutinasi inhibisi, dan dianalisa menggunakan uji one-way ANOVA dengan uji lanjut Duncan (α:0,05). Hasil penelitian menunjukkan bahwa terjadi imunosupresi pada kelompok yang ditantang dengan virus IBD (K1 dan K3). Meskipun demikian, titer antibodi ND yang dihasilkan oleh kelompok yang divaksin IBD-inaktif (K1) mampu memproduksi antibodi pada tingkat protektif dengan nilai ≥ log 23 selama masa pengamatan. Berdasarkan hasil tersebut disimpulkan bahwa vaksinasi IBD inaktif mampu melindungi ayam dari efek imunosupresi akibat infeksi virus IBD.

ABSTRACT

The study was aimed to determine the profile of immune response against Newcastle Disease in broiler that were vaccinated with IBD-inactivated vaccine and challenged IBD virus. Two hundred Cobb broiler chickens were divided into four groups (K1, K2, K3 and K4). On the 4th day, K1 and K2 groups were vaccinated with ND active vaccine and ITA ND+IBD inactivated vaccine, while K3 and K4 groups were vaccinated with ND active and ND inactivated vaccine. Challenge test with IBD virus was performed on the 14th day. Revaccination of Newcastle Disease for all groups was carried out on the 28th day. Blood sample for antibody measurements was drawn on the 4th, 14th, 21st, 28th and 42nd day. Measurements of ND antibodies were done by haemaglutination inhibition test (HI), and the analysis used one-way test of ANOVA by Duncan test advanced Multiple Range Test (α: 0.05). The results showed that the effect of immunosuppressions were occured in the groups which was challenged by IBD virus (K1 and K3). However, the group of IBD-inactivated vaccinated (K1) were able to produce ND protective antibody ≥ log 23 during the period of observation. Based on these results we can concluded that vaccination with IBD-inactivated vaccine could protect chickens from the effects of immunosuppression due to viral infection IBD.
