ABSTRAK

Penelitian ini dilakukan untuk mengetahui aktivitas ekstrak air pegagan (Centella asiatica) terhadap tikus putih jantan strain Wistar yang diinduksi parasetamol. Tikus dibagi dalam lima kelompok perlakuan yaitu kelompok kontrol (akuades) dan empat kelompok formula ekstrak air pegagan (F 6, F 10, F 12, and F 16). Tikus diinduksi dengan parasetamol dosis 1000 mg/kg bw (p.o) sebagai dosis toksik untuk menginduksi kerusakan hati. Formula ekstrak air pegagan diberikan setelah satu jam induksi parasetamol dengan dosis 1500 mg/kg bw (p.o). Perlakuan diberikan selama 8 hari dan darah diambil sebanyak tiga kali (sebelum perlakuan, sesudah pemberian parasetamol, dan akhir perlakuan) dari arteri pada ekor tikus dan dianalisis kadar SGPTnya. Kerusakan hati yang dihasilkan oleh pemberian parasetamol berhasil dihambat oleh kelompok F 16 dengan bukti berupa penurunan level kadar SGPT. Aktivitas hepatoprotektor ekstrak air pegagan juga didukung oleh pemeriksaan secara histopatologi.

ABSTRACT

This study was undertaken to investigate activity of Centella asiatica extract against paracetamol induced liver damage in male Wistar albino rats. Rats were divided into five groups namely control (aquadest), herb extract with four formulations which were F 6, F 10, F 12, and F 16. Rats were administered paracetamol at dose 1000 mg/kg bw (p.o) as toxic dose for inducing hepatotoxicity. Herb extract in each formula was given at dose 1500 mg/kg bw (p.o) one hour after paracetamol administration. The treatment was given for eight days and blood was collected three times (before treatment, after paracetamol treatment, and last treatment) from tail artery and analysed for serum glutamate pyruvate transminase (SGPT). The hepatoxicity produced by acute paracetamol administration was found to be inhibited by Centella asiatica at group F 16 with evidence of decreased level of serum glutamate pyruvate transminase (SGPT) consentration. The hepatoprotective activity Centella asiatica was also supported by histopathological studies of hepatocytes.
